Hydroxypropyl beta-cyclodextrin is a ring-shaped molecule made from starch. The inside of the ring repels water and the outside attracts it, so it can hold a drug that does not dissolve in water inside the ring and carry it into solution. That makes it possible to turn such drugs into injections, eye drops and oral solutions. It is also the odour-trapping ingredient in some fabric fresheners.
Technical details
Hydroxypropyl beta-cyclodextrin is a derivative of beta-cyclodextrin, a ring of seven glucose units produced enzymatically from starch. The ring's cavity is hydrophobic and its exterior hydrophilic, so it forms inclusion complexes with poorly soluble molecules and carries them into aqueous solution. Hydroxypropylation disrupts the hydrogen bonding that limits the parent cyclodextrin to about 18 g per litre, raising solubility above 500 g per litre and making the material suitable for parenteral use. The degree of molar substitution changes complexation behaviour, so it is fixed in the specification. Ehlinar supplies parenteral, oral and technical grades.
Appearance: White to off-white amorphous powder; very soluble in water, above 50% w/v.

Frequently asked questions
- Why not use plain beta-cyclodextrin?
- Beta-cyclodextrin itself dissolves only to about 18 g per litre and is not suited to injection. The hydroxypropyl derivative is far more soluble and is the form used in parenteral products.
- What does molar substitution mean for my formulation?
- It is the average number of hydroxypropyl groups per glucose unit. It affects solubility and how strongly a given drug is complexed, so a formulation developed on one substitution range should stay on it.
- Which grade for injection?
- The pharmacopoeial parenteral grade, with low endotoxin and the documentation a regulated filing needs. Tell us the pharmacopoeia and the substitution range when you ask for a quote.
