D-Mannitol is a sugar alcohol found naturally in mushrooms, seaweed and many fruits. Medicine makers use it to give freeze-dried injections a firm cake that dissolves quickly, and as a filler in tablets, because it tastes sweet and cool and does not absorb moisture. It is also a sweetener in sugar-free sweets and chewing gum, and hospitals use it as an infusion to reduce swelling in the brain.
Technical details
D-Mannitol is a six-carbon sugar alcohol, a stereoisomer of sorbitol, produced by hydrogenating fructose-rich sugars or extracted from seaweed. In freeze-drying it crystallises during freezing and forms a mechanically stable cake, so it is paired with an amorphous stabiliser: trehalose or sucrose protects the protein, and mannitol gives the cake its structure. It is non-reducing and non-hygroscopic, so it does not take part in Maillard reactions with amine-bearing drugs and does not draw moisture into a tablet. Ehlinar supplies pharmacopoeial, parenteral and food grades.
Appearance: White crystalline powder or granules; odourless, with a sweet, cooling taste; freely soluble in water; not hygroscopic.

Frequently asked questions
- Why is mannitol used together with trehalose in freeze-drying?
- They do different jobs. Trehalose stays amorphous and protects the protein; mannitol crystallises and gives the dried cake strength and a clean appearance. Used alone, mannitol can crystallise away from the protein and leave it unprotected, which is why the two are combined.
- Which grade for tablets?
- Spray-dried or granular grades for direct compression; fine powder grades for wet granulation. For injectables and freeze-drying, specify the low-endotoxin parenteral grade.
- Is mannitol the same as sorbitol?
- No. They are stereoisomers with the same formula, but sorbitol is hygroscopic and mannitol is not, so they behave very differently in tablets and dry products. They have different CAS numbers.
